Succinylation of the collagen was performed by the dropwise addition of 5 ml of 2% succinic anhydride in acetone to 200 ml of 0.1% isinglass in 0.05% acetic acid (Wang et al., 1978). The pH was maintained at 9 by the addition of NaOH. Succinylated collagen was precipitated by adjustment of the pH to 4.2 with HCl, and the precipitate was collected by centrifugation (4000×g for 10 min at 4°C). The pellet was washed three times in dilute HCl solution pH 4.2 and finally resuspended in 0.01% malic acid. Succinylation of the collagen was confirmed by iso-electric focusing (not shown).
