Molecular Physiology lab focuses on research related to the role of glyoxal (GO) and methylglyoxal (MGO) in cellular physiology and disease. Glyoxals are known to be produced as a by-product of glucose metabolism, which are toxic to nucleotides and proteins. Glyoxals react non-enzymatically with amino groups of proteins, forming advanced glyocation end products (AGEs), which were implicated in aging, diabetes, and neurodegenerative diseases, as well as in neuronal apoptosis. Our research includes screening of genes related to glyoxal metabolism and characterizing their functions to elucidate molecular mechanisms involving glyoxals. Detoxification of glyoxals involves NADPH and GSH. Using various model systems, we found that Parkinson’s disease associated DJ-1 is a novel type of glyoxalase, detoxifying glyoxals in the absence of cofactor.
