Research aim: The chronic inflammation is now being a great deal of attention due to being primary causes of cancer and cardiovascular disease. However, the fundamental principle with respect to molecular mechanism of the inflammatory response is largely unknown. The inflammatory response includes a variety of cellular communications and internal signaling mediators, which results in complexity in signaling networks. Nonetheless, we have looked into the intracellular inflammatory signaling transduction in depth, and we have drawn a fundamental signaling frame; that is, intracellular inflammatory response constitutes two major signaling axes: Reactive oxygen species and NF-kB. By understanding the molecular networks surrounding these axes, we hope to establish a key inflammatory signaling network. 1. To elucidate the mode of ROS action as signaling messenger via studying the function of antioxidant enzymes in a variety of signaling pathways, which include growth factors and cytokines, for example, platelet-derived growth factor and tumor necrosis factor-a. 2. To discover a fine signal tuner for regulating the activation of nuclear factor-kB (NF-kB), the most important pro-inflammatory transcriptional factor.
