과산화 수소를 이용한 균주 선별법

UV돌연변이체 중 활성산소에 저항성을 가진 균주를 선별하려고 합니다. 그래서 과산화수소를 사용하려고 하는데, 아시다시피 과산화수소는 액체이고, 또한 배지에 섞어면 날라가 버릴 것 같은데 좋은 방법이 없나 싶어 이렇게 글 올립니다. 혹시 이와 관련된 논문이나 방법 하시는 분 조언 좀 부탁드빈다.

    논문을 보던 중 질문하신 내용이 있는 것 같아 퍼왔습니다. 참고해 보세요. Oxidative stress assays. Disc inhibition assays were used to test sensitivity to hydrogen peroxide and cumene hydroperoxide. Bacteria were grown in LB broth overnight and adjusted to 108 c.f.u. ml21 in LB broth. One millilitre of this bacterial suspension was added to 9 ml molten LB agar, mixed, and dispensed into Petri plates. Following solidification of agar, paper discs (6 mm) were soaked with 6 ml of 30% (v/v) hydrogen peroxide or 15 ml of 10% (w/v) cumene hydroperoxide and placed on the surface of agar plates. To test for sensitivity to reactive nitrogen intermediates (RNI), bacteria were grown in LB broth (pH 5) containing sodium nitrite for 16 h at 37 uC. Following incubation, cultures were diluted and plated onto LB agar quadrant plates for viable counts.
