    Structural Studies on the Specificity of the CML Drug STI-571/Gleevec

    John Kuriyan is interested in the construction, mechanism, and regulation of the molecular machines and switching devices that carry out signal transduction and DNA replication. His laboratory uses x-ray crystallography to determine the three-dimensional structures of the component proteins, along with biochemical and biophysical analyses to elucidate mechanisms.

    The concept of personalized medicine ??? tailoring health care to an individual???s genetic profile - represents a central promise of the Human Genome Project. Genetic information is already used to guide the management of some health risks. However, critics have questioned whether the model of personalized medicine can serve as the basis for broad changes in health care, citing inadequate evidence and theoretical limitations in the predictive value of genetic information. Over time, many evidence gaps will be closed. Important questions will remain: What level of risk is sufficient for health care action? What resources are needed for an effective response to genetic risk? Addressing these questions will require attention to the personal and societal values that inform preventive health care.
